SRX5399538: Shotgun sequencing of Lecythidaceae: leaf
1 ILLUMINA (Illumina HiSeq 2000) run: 19.8M spots, 3G bases, 2.1Gb downloads

Design: ilica-dried leaf tissue from herbarium-vouchered collections was collected by Scott Mori and colleagues and loaned by the New York Botanical Garden. Total genomic DNA was extracted from 20 mg of dried leaf tissue using the NucleoSpin Plant II extraction kit (Machery-Nagel, Bethlehem, Pennsylvania, USA) with SDS lysis buffer. Prior to DNA library preparation, 5 g of total DNA was fragmented using a Covaris S-series sonicator (Covaris Inc., Woburn, Massachusetts, USA) following the manufacturers protocol to obtain approximately 300-bp insert sizes. We prepared the sequencing library using the NEBNext DNA library Prep Master Mix and Multiplex Oligos for Illumina Sets (New England BioLabs Inc., Ipswich, Massachusetts, USA) according to the manufacturers protocol. Size selection was carried out prior to PCR using Pippin Prep (Sage Science, Beverly, Massachusetts, USA)
